Considered as an employee, I was familiarized with general Quality Consultancy and
Certification Protocols
* Documentation (prepared the Quality Management System Manual, the basic forms and
the mandatory procedures according to ISO 9001:2008 standards for various companies)
* Conducted Implementation, Internal as well as Certification audits for several companies
across UAE
* Through auditing, I also got the opportunity to learn about several industries and their
associated works
* Attended and Assisted employees for quality training sessions to various firms
As a trainee, I was asked to observe the various tests performed in the pathology lab and I
was put on rotations in different fields such as microbiology, biochemistry,
histology/pathology, general blood work
* Learnt about the instrumentation utilized in the lab
* Made an inventory of the different instruments whilst particularly noting some specifications
such as the Installation Qualification, Operational Qualification, and Performance
Qualification. These are studied for the validation of the equipment
* Had to crosscheck the Standard Operating Procedure (SOP) of the instrument with the
actual method used by the pathologist
* Made an inventory of the all the tests done in the lab as well as those that are outsourced
along with appropriate details
* Learnt and presented about the chemical safety in the lab
* Reorganized the lab to prepare it for the forthcoming accreditation

LAB ROTATIONS:
Dr. Richard .L. Eckert's Lab: Working for a period of two months, I focused on deciphering the
location and key role of three genes in human keratinocytes (TG2, EZH2 and p63). To identify
location, I performed IHC on paraffinized human adult and fetal skin sections. To study the
regulation and interplay among these genes, EZH2 silencing was done using siRNA and
electroporation technique. The confirmation and consequence of this gene silencing was
confirmed through western blotting.
Dr. Ricardo .A. Feldman's Lab: Started with (Gaucher iPSC derived) Mesenchymal Stem Cell
differentiation into osteoblasts. I was familiarized with lab techniques such as Imunofluorescence,
Western Blotting, Immunohistochemistry & Cell Culture.
THESIS PROJECT:
Advisor: Dr. Ricardo .A. Feldman
Project Aim: To study the altered WNT signaling in osteoblasts derived from Gaucher iPSC’s
Firstly, I was trained to handle the various MSC cell lines and to differentiate them into
osteoblasts. Once the osteoblast formation and characterization was done, our hypothesis was
that the mutant osteoblasts display an aberrant WNT signaling.
I utilized techniques such as Immunofluorescence and western blotting to show that the mutant
osteoblasts had reduced β-catenin levels which concur with their observed defectiveness (such
as decreased RUNX2 and Cbfa1expression, Alkaline Phosphatase and Alizarin Red Staining and
lower secretion of collagen 1 as compared to the control).
This was due to an increase in GSK-3β levels (GSK-3 β is known to phosphorylate and thus mark β-
catenin for degradation).
The reduced inactive (phosphorylated at Ser9) GSK-3β levels suggest that AKT regulation might
be involved as well.
Treatment with rGCase reversed the effects almost entirely in all the cases.
