Assists in the maintenance and propagation of rootcrops planting materials
Wash laboratory glasswares, tools and equipments. Sterilize on autoclave 2-3 hours at 15 psi. After which, it will be transferred in the oven for dry sterilization.
Prepare media for culturing in-vitro potato plantlets. Dispense it on test tubes. Plug. Then sterilize it on autoclave for 15-30 minutes at 15 psi.
Sterilize media will be brought in the inoculation room for cooling. Ultraviolet lamp will be opened overnight for sterilizing the laminar flow.
On the following day, before entering the room, hands will be washed and then wear laboratory gown and gloves.
Before starting to subculture, the laminar flow will be disinfected with 70% alcohol/95 % ethyl alcohol. Spray also the test tubes and plugs with 95% ethyl alcohol to remove extra dirt attach.
Aseptically, use sterilized forceps to pull out the plantlet. Then cut in between each node with sterilized scissor. Lamps are beside the working area to move away organism that floating in the air that can cause contamination.
Place 5 cut nodes on each test tube and plugged. Then repeat with the same procedure. A lamp is always placed inside the laminar flow in the middle while subculturing.
Discard contaminated cultures on bottles and test tubes, sterilize and wash. Then soak glasswares on trays with chlorox and detergent soap. The glasswares will be wash on the following day. Washed glasswares will be sterilized again on the autoclave for specified time. After which, the glasswares will be transferred again on oven for dry sterilization.
Assist in the conduct of RDE on rootcrops management (i.e prepare data, gathering of tools and other instrument in the conduct of RDE activities, application of treatments, etc).
Assists on stem-cutting on potato production.
Does other activities as maybe required by the supervisor.
