Dr. Ram Manohar Lohia Avadh University, Faizabad, Faizabad-224001, Uttar Pradesh, India. ❖ BRIEF SUMMARY OF THE WORK DONE DURING Ph.D. Since pullulan is an important polysaccharide with 300 patents of its applications in various industries, one of the most promising use for the production of biodegradable plastics. Till date only mesophilic strains of A. pullulans has been exploited for production of this polysaccharide worldwide while the thermotolerant strain is the need of hour. In this context we have successfully isolated a thermotolerant higher pullulan producing strain from natural ecosystem. This fungus has been critically evaluated at different physico-chemical and nutritional parameters for pullulan production in flask, stirred and non-stirred fermentation condition in a fermentor of 7 liter capacity. A suitable mutant has also been successfully developed for higher pullulan without disturbing thermotolerant nature of wild strain. The review of literature clearly indicates that less attention has been paid towards development of bioreactor in order to achieve continuous production. Therefore, a novel fermentation model having immobilized microbial cells of A. pullulans has been successfully developed in order to achieve continuous production of pullulan. This study has been compared in several designs with all correction factors like backflow of air, viscosity, dilution rate and other factors to achieve the constant production of pullulan. In this investigation a novel fungus Eurotium chevalieri has been isolated and optimized at various physico-chemical and nutritional parameters, a novel aspect, which has been reported first time for higher pullulan which is at par to the production of pullulan with A. pullulans.
