1) Isolate and make cultures of bacteria or other microorganisms in prescribed media, controlling moisture, aeration, temperature, and nutrition.
2) Perform tests on water, food and the environment to detect harmful microorganisms and to obtain information about sources of pollution and contamination.
3) Examine physiological, morphological, and cultural characteristics, using microscope, to identify and classify microorganisms in human, water, and food specimens.
4) Provide laboratory services for health departments, for community environmental health programs and for physicians needing information for diagnosis and treatment.
5) Observe action of microorganisms upon living tissues of plants, higher animals, and other microorganisms, and on dead organic matter.
6) Investigate the relationship between organisms and disease, including the control of epidemics and the effects of antibiotics on microorganisms.

The aim of this work was to evaluate the presence of classical bacterial indicators of water pollution as well as free-living amoebae in different surface water sources in Sharkyia governorate. Culture methods were used to detect bacterial indicators, free-living amoebae and some pathogenic bacteria namely Bacillus subtilis, E. coli O157:H7, Pseudomonas aeruginosa and Staphylococcus epidermidis. Furthermore, the predominant isolates of bacteria were confirmed by using BilogMicroStation™ system/MicroLog Version 5.1.1. The bactericidal activity of chlorine on the identified bacterial strains was investigated. The interaction between Acanthamoeba quina and bacterial strains (E. coli O157:H7, Pseudomonas aeruginosa, Bacillus subtilis and Staphylococcus epidermidis) revealed that the use of Gram positive bacterium (Staphylococcus epidermidis) gave the highest yield of Acanthamoeba quina, followed by Gram negative bacteria (Pseudomonas aeruginosa and E. coli), while Bacillus subtilis was found consumed less by amoebae and consequently elicited a low to moderate trophozoite growth. the use of chlorine for inactivation of the isolated strains of bacteria revealed that the removal percentage of E. coli O157:H7, Staphylococcus epidermidis, Pseudomonas aeruginosa and Bacillus subtilis were 99.98, 100, 99.92 and 98.83% respectively
