During this Period I work as Researcher in Project which is Detection of Human Papilloma Viruses Type 16 in Sudanese Women Patients with Cervical Carcinoma using Molecular Biology Techniques. I collect 158 of Paraffin Embedded Tissues Blocks from Sudanese Women Patients with Cervical Carcinoma such as malignant or benign from Omdurman Maternity Hospital and National Laboratories of Health. I collect their ages, Clinical information, and degree of cancer differentiation from Hospitals records. I isolate Genomic DNA from Paraffin Embedded Tissues using Guanidine Chloride manual method after sectioning samples using Microtome and de paraffinaization the Blocks using Incubater (heating) and Protienase k. The quality of Genomic DNA measure by using Nano drop. Then, I detect HPV16 in DNA using HPV16 and18 Florescence Diagnostic Kits in some samples to used lately as positive control sample to optimize conventional PCR condition. The positive control samples and remaining sample with different concentration and differents programs of Gradient and Touch down PCR with HPV16 E6 specific primer provide no result in gel electrophoresis at predicted size. So, I use Real time PCR with SYBER GREEN Kit and HPV16 E6 specific primer showing the presence of HPV16 in samples is 10% of Malignant cases and 6% in benign - Lab of Physiology and Molecular Biology+Lab of Leishmania-Department of Zoology, Faculty of Science, University of Khartoum- Department of Virology, Central Lab- Khartoum.
