At fertilization, an increase of intracellular calcium is required for egg activation and begins subsequent embryonic development. Inositol 1, 4, 5- trisphosphate (IP3), the second messenger of phospholipase Cγ, regulates the initial calcium rise from egg’s endoplasmic reticulum via activation of IP3 receptors. However, the mechanism of how PLCγ interacts with other proteins to trigger egg activation is unclear. SH2 domain of PLCγ is assumed to involve into an important process of signaling network for the beginning of egg activation by binding phosphorylated tyrosine residues. To examine the proteins that have an interaction with SH2 domain of PLCγ during fertilization, GST-SH2 fusion protein was used to inject into the starfish Asterina miniata’s egg. Therefore, SH2 domain was subcloned into a pGEX vector to make a GST-SH2 fusion protein in order to do affinity interaction to get some egg’s proteins interact with the SH2 domain.
